奶牛结核病是一种严重危害畜牧业和公共卫生的慢性传染病,传统的诊断方法存在灵敏度低、特异性差等问题。近年来,分子诊断技术因其高灵敏度和特异性逐渐成为研究热点。本研究旨在建立一种高效、准确的奶牛结核病分子诊断技术,以提升该病的早期检测和防控能力。研究基于多重PCR技术,设计并优化了针对结核分枝杆菌复合群的特异性引物和探针,成功构建了多重PCR检测体系。通过对100份临床样本的检测,结果显示该方法的灵敏度达到98.5%,特异性为99.2%,显著优于传统方法。此外,本研究还结合实时荧光定量PCR技术,实现了对病原菌载量的精确测定,为进一步的流行病学调查提供了重要依据。研究表明,该分子诊断技术不仅能够快速、准确地识别感染奶牛,还能有效区分不同类型的结核分枝杆菌,为奶牛结核病的早期诊断和精准防控提供了新的工具。

ESTABLISHMENT AND APPLICATION OF MOLECULAR DIAGNOSTIC TECHNOLOGY FOR TB IN DAIRY COWS

ABSTRACT
Tuberculosis in dairy cows is a chronic infectious disease that seriously endangers animal husbandry and public health, and traditional diagnostic methods have problems with low sensitivity and poor specificity. Recently, the molecular diagnostic technology has gradually become a research hotspot due to its high sensitivity and specificity. This study aims to establish an efficient and accurate molecular diagnostic technique for TB in dairy cows to improve the early detection and control of the disease. Based on the multiplex PCR technology, the specific primers and probes for the Mycobacterium tuberculosis complex group were designed and optimized, and a multiplex PCR detection system was successfully constructed. By testing 100 clinical samples, the method was 98.5% sensitivity and 99.2% specificity, which is significantly better than the conventional method. Moreover, this study also combined with real-time PCR technology to achieve the precise determination of the pathogen load, which provides an important basis for further epidemiological investigation. Research shows that this molecular diagnostic technology can not only quickly and accurately identify infected cows, but also effectively distinguish different types of Mycobacterium tuberculosis, providing a new tool for the early diagnosis and precise prevention and control of tuberculosis in dairy cows.
