槐米中芦丁的提取工艺及抗氧化活性评价
摘 要
槐米作为传统中药材,其内含丰富的生物活性成分,尤其是芦丁,这是一种具有广泛药理活性的黄酮类化合物。芦丁不仅具有抗炎、抗菌、抗病毒等生物活性,还展现出强大的抗氧化能力,对于预防和治疗多种慢性疾病具有重要意义。本研究针对槐米中芦丁的提取工艺进行了系统优化,对比了不同提取方法对芦丁得率的影响,并通过实验设计优化了提取工艺参数,最终确定了最佳提取工艺条件并进行了验证。在此基础上,采用高效液相色谱法对芦丁进行了分离纯化,并利用多种光谱手段对芦丁的结构进行了表征与分析。为全面评价槐米中芦丁的抗氧化活性,本研究采用了DPPH自由基清除能力测定、ABTS自由基清除能力测定以及FRAP铁离子还原能力测定三种方法。优化后的提取工艺能显著提高芦丁的得率与纯度,且槐米中芦丁展现出强大的抗氧化活性。

EXTRACTION PROCESS AND EVALUATION OF ANTIOXIDANT ACTIVITY IN LOCUST RICE
ABSTRACT
As a traditional Chinese medicine, locust rice contains rich bioactive ingredients, especially rutin, which is a kind of flavonoids with extensive pharmacological activities. Rutin not only has anti-inflammatory, antibacterial, antiviral and other biological activities, but also shows a strong antioxidant capacity, which is of great significance for the prevention and treatment of a variety of chronic diseases. In this study, the extraction process of rutin in locust rice was systematically optimized, and the influence of different extraction methods on the yield of rutin was compared, and the extraction process parameters were optimized through experimental design, and the best extraction process conditions were determined and verified. Furthermore, rutin was isolated and purified by HPLC, and its structure was characterized and analyzed by various spectral means. To comprehensively evaluate the antioxidant activity of rutin in locust rice, three methods were DPPH radical scavenging, ABTS radical scavenging and FRAP. The optimized extraction process can significantly improve the yield and purity of rutin, and rutin shows strong antioxidant activity in locust rice.
