Extraction and Separation of Active Ingredients from Traditional Chinese Medicine Compound Formulas and Their Pharmacological Studies 

Abstract
Traditional Chinese medicine (TCM) formulas, as a core component of TCM science, provide significant insights for modern drug development through their multi-component and multi-target synergistic mechanisms; however, the extraction and separation of active ingredients as well as pharmacological evaluation remain critical challenges in this field. This study focuses on classical TCM formulas, aiming to optimize extraction and separation techniques combined with systematic pharmacological analysis to elucidate their active components and mechanisms of action. Supercritical CO2 extraction coupled with high-performance liquid chromatography was employed to achieve precise extraction and quantitative analysis of major active ingredients in the formula, while in vitro and in vivo experiments validated its pharmacological effects in anti-inflammation, antioxidation, and immune regulation. The results indicate that specific flavonoid and alkaloid compounds in the formula exhibit significant synergistic effects, potentially involving the modulation of the NF-κB signaling pathway and the Nrf2 antioxidant system. Additionally, this study innovatively incorporates network pharmacology to construct an “ingredient-target-pathway” association network, further clarifying the molecular basis of multi-component synergy in the formula. These findings not only establish a theoretical foundation for the modernization of this formula but also offer new technical approaches and scientific evidence for TCM formula research. Overall, by integrating modern analytical technologies and pharmacological evaluation systems, this study successfully deciphers the active ingredients and mechanisms of action of TCM formulas, making a positive contribution to the advancement of TCM modernization and internationalization.
