川芎作为传统中药在心血管疾病治疗中具有重要地位,但其具体药理机制尚未完全阐明。本研究旨在系统探讨川芎主要活性成分及其对心脑血管疾病的治疗作用机制。采用高效液相色谱-质谱联用技术对川芎提取物进行成分分析,并通过体外细胞实验和动物模型评估其药理活性。研究结果表明,川芎中主要含有阿魏酸、藁本内酯等活性成分,这些成分通过抑制炎症因子释放、调节血管内皮功能、改善微循环等多重途径发挥治疗作用。其中,阿魏酸显著降低TNF-α和IL-6水平,藁本内酯可有效抑制血小板聚集。动物实验显示,川芎提取物能显著改善心肌缺血再灌注损伤模型大鼠的心功能指标,并减轻脑缺血后的神经功能缺损评分。本研究首次系统阐明了川芎多组分协同作用的分子机制,证实其通过PI3K/Akt和MAPK信号通路调控细胞凋亡和炎症反应。研究结果为川芎在心脑血管疾病中的临床应用提供了科学依据,并为开发新型多靶点药物奠定了理论基础。

PHARMACOLOGICAL BASIS OF LIGUSTICUM XIONG IN THE TREATMENT OF CARDIOVASCULAR AND CEREBROVASCULAR DISEASES

ABSTRACT
As a traditional Chinese medicine, Chuanxiong plays an important role in the treatment of cardiovascular diseases, but its specific pharmacological mechanism has not been fully elucidated. This study aimed to systematically investigate the main active components of Chuanxiong and its therapeutic mechanism on cardiovascular and cerebrovascular diseases. Constitupositional analysis of C. chuanxiong extract was performed by HPLC-MS, and its pharmacological activity was assessed by in vitro cell experiments and animal models. The results show that Ligusticum chuanxiong mainly contains active ingredients such as ferulic acid and chaff lactone, which play a therapeutic role in multiple ways such as inhibiting the release of inflammatory factors, regulating vascular endothelial function and improving microcirculation. Among them, ferulic acid significantly reduced the levels of TNF- α and IL-6, and lacoltone could effectively inhibit platelet aggregation. Animal experiments showed that Ligusticum extract could significantly improve the cardiac function index of myocardial ischemia and reperfusion injury model rats and reduced the neurological deficit score after cerebral ischemia. This study was the first to systematically elucidate the molecular mechanism of Chuanxiong multicomponent synergy and confirmed its regulation of apoptosis and inflammation through PI3K / Akt and MAPK signaling pathways. The results provide a scientific basis for the clinical application of Chuanxiong in cardiovascular and cerebrovascular diseases, and lay a theoretical foundation for the development of new multi-target drugs.
