金银花提取物对流感病毒抑制效果的体外研究

摘  要

本研究旨在探讨金银花提取物对流感病毒的体外抑制作用及其潜在机制。鉴于流感病毒对人类健康的持续威胁,以及传统中药在抗病毒领域的应用价值,本研究选取具有广泛药用价值的金银花作为研究对象。采用水提法制备金银花提取物,通过细胞病变效应抑制实验、空斑减少实验和实时荧光定量PCR等方法,系统评估了提取物对甲型流感病毒H1N1的抑制作用。结果显示,金银花提取物在非细胞毒性浓度下显著抑制H1N1病毒的复制,其半数抑制浓度(IC50)为12.5 μg/mL,治疗指数(TI)达到32。进一步研究发现,提取物能够下调病毒核蛋白(NP)和基质蛋白(M1)的表达水平,并抑制病毒诱导的炎症因子IL-6和TNF-α的过度释放。机制研究表明,金银花提取物可能通过干扰病毒吸附和进入宿主细胞的过程发挥抗病毒作用。

STUDY ON INHIBITORY EFFECT OF EXTRACT OF HONEYSUCKLE ON INFLUENZA VIRUS IN VITRO

ABSTRACT

The aim of this study was to investigate the inhibitory effect of honeysuckle extract on influenza virus in vitro and its potential mechanism. In view of the continuous threat of influenza virus to human health and the application value of traditional Chinese medicine in the field of antiviral, this study selected honeysuckle, which has a wide range of medicinal value, as the research ob ject. The antibacterial activity of the extract against influenza A (H1N1) virus was evaluated by cytopathic inhibition assay, plaque reduction assay and real-time fluorescence quantitative PCR. The results showed that the extract of honeysuckle significantly inhibited the replication of H1N1 virus at non-cytotoxic concentration, with a median inhibitory concentration (IC50) of 12.5 μg/mL and a therapeutic index (TI) of 32. Further studies showed that the extract down-regulated the ex pression levels of viral nucleoprotein (NP) and matrix protein (M1), and inhibited the excessive release of virus-induced inflammatory cytokines IL-6 and TNF-α. Mechanism studies suggest that honeysuckle extract may play an antiviral role by interfering with the process of viral adsorption and entry into host cells.
