发光金纳米粒子的酸性微环境响应调控及其成像研究

摘    要

  发光金纳米粒子在生物成像、疾病诊断和治疗监测等领域展现出巨大潜力,酸性微环境广泛存在于肿瘤组织等病理区域,针对此特性进行响应调控可提高成像的特异性和灵敏度。本研究旨在构建一种对酸性微环境具有响应性的发光金纳米粒子,并对其成像性能进行深入探究。通过合理设计合成路线,在金纳米粒子表面修饰特定的功能基团,使其荧光强度随pH值降低而显著增强。采用紫外 - 可见吸收光谱、透射电子显微镜等手段表征了粒子的形貌与光学性质,利用细胞实验考察其生物相容性及细胞摄取情况,结果显示该粒子具有良好的生物相容性且能被细胞有效摄取。进一步以肿瘤细胞为模型,验证了其在酸性环境下荧光增强效果明显,可实现对肿瘤细胞的高效识别与成像。创新点在于巧妙地将酸性微环境响应特性赋予金纳米粒子,拓展了其在生物医学领域的应用范围,为开发新型智能成像探针提供了新思路,主要贡献是为肿瘤等疾病的早期诊断和精准医疗奠定了基础。

Abstract 
  Luminescent gold nanoparticles exhibit significant potential in biological imaging, disease diagnosis, and therapeutic monitoring. Given that acidic microenvironments are prevalent in pathological regions such as tumor tissues, responsive regulation targeting this characteristic can enhance the specificity and sensitivity of imaging. This study aims to construct luminescent gold nanoparticles with responsiveness to acidic microenvironments and to thoroughly investigate their imaging performance. By rationally designing the synthetic route, specific functional groups were modified on the surface of gold nanoparticles, resulting in a notable increase in fluorescence intensity as pH decreases. The morphology and optical properties of the particles were characterized using techniques such as UV-visible absorption spectroscopy and transmission electron microscopy. Cellular experiments were conducted to evaluate their biocompatibility and cellular uptake, revealing excellent biocompatibility and efficient cellular internalization. Further validation using tumor cells as a model demonstrated a significant enhancement in fluorescence under acidic conditions, enabling effective recognition and imaging of tumor cells. The innovation lies in ingeniously endowing gold nanoparticles with acidic microenvironment-responsive characteristics, thereby expanding their applications in biomedical fields and providing new insights for developing intelligent imaging probes. The primary contribution is laying the foundation for early diagnosis and precision medicine in diseases such as cancer.
