摘 要
口腔颌面部肿瘤是临床常见的恶性肿瘤,具有侵袭性强、易复发转移等特点,传统治疗方法如手术、放疗和化疗虽有一定疗效但存在诸多局限性。近年来,基因治疗作为一种新兴的治疗手段为口腔颌面部肿瘤的治疗带来了新的希望。本研究旨在探讨基因治疗在口腔颌面部肿瘤中的应用进展,通过查阅大量国内外相关文献资料并结合实验研究,对基因治疗的多种策略进行综述。研究表明,基因治疗可通过多种机制发挥抗肿瘤作用,如自杀基因疗法可使肿瘤细胞特异性表达有毒物质从而杀伤肿瘤细胞;RNA干扰技术能够抑制肿瘤相关基因的表达;免疫基因治疗则增强机体对肿瘤细胞的免疫应答。创新点在于整合多种基因治疗方法的优势,提出联合治疗的新模式,并且深入分析了不同基因治疗策略的安全性和有效性,在动物模型中验证了部分联合方案的有效性,为进一步开展临床试验提供了理论依据。结论是基因治疗为口腔颌面部肿瘤的治疗开辟了新途径,尽管仍面临一些挑战,但其潜在的巨大价值不可忽视,未来有望成为一种重要的治疗方式。

Abstract
Oral and maxillofacial tumors are clinically common malignant neoplasms characterized by strong invasiveness and a tendency for recurrence and me tastasis. Conventional treatments such as surgery, radiotherapy, and chemotherapy have shown certain efficacy but are limited by various constraints. In recent years, gene therapy has emerged as a promising therapeutic approach, offering new hope for the treatment of oral and maxillofacial tumors. This study aims to review the advancements in the application of gene therapy for these tumors by examining extensive domestic and international literature and integrating experimental research. Gene therapy can exert antitumor effects through multiple mechanisms: suicide gene therapy enables tumor cells to specifically express toxic substances, thereby killing them; RNA interference technology inhibits the ex pression of tumor-related genes; and immunogene therapy enhances the body's immune response to tumor cells. An innovative aspect of this research is the integration of the advantages of various gene therapy methods, proposing a combined treatment model. Furthermore, it provides an in-depth analysis of the safety and effectiveness of different gene therapy strategies, validating some combination regimens in animal models, thus providing theoretical support for future clinical trials. The conclusion is that gene therapy opens up new avenues for the treatment of oral and maxillofacial tumors. Although challenges remain, its significant potential value cannot be overlooked, and it is expected to become an important therapeutic modality in the future.
