摘 要
抑郁症是一种复杂的神经精神疾病,其高发病率和低治愈率促使人们寻找更安全有效的治疗策略。本研究旨在筛选具有抗抑郁作用的中药活性成分并探讨其作用机理,为新型抗抑郁药物开发提供理论依据。通过建立基于网络药理学和分子对接技术的综合筛选平台,结合细胞实验和动物模型验证,我们从多种传统中药中筛选出若干潜在活性成分,如人参皂苷Rg1、黄芩苷等,并揭示其可能通过调节5-羟色胺系统、炎症通路及神经可塑性相关基因发挥抗抑郁作用。研究结果表明,这些活性成分能够显著改善抑郁样行为,并在分子水平上调控关键靶点如5-HT1A受体、BDNF及NF-κB信号通路。本研究创新性地整合多组学数据与实验验证,明确了中药抗抑郁作用的多靶点协同机制,为中医药现代化研究提供了新思路,同时为开发高效低毒的抗抑郁药物奠定了基础。关键词:抑郁症; 中药活性成分; 网络药理学; 分子对接; 5-羟色胺系统
Abstract
Depression is a complex neuropsychiatric disorder, and its high incidence and low cure rate have prompted the search for safer and more effective treatment strategies. This study aims to screen active components of traditional Chinese medicine with antidepressant effects and explore their mechanisms of action, providing theoretical support for the development of novel antidepressant drugs. By establishing an integrated screening platform based on network pharmacology and molecular docking technology, combined with cellular experiments and animal model validations, we identified several potential active components from various traditional Chinese medicines, such as ginsenoside Rg1 and baicalin, and revealed that they may exert antidepressant effects by modulating the 5-hydroxytryptamine system, inflammatory pathways, and genes related to neuroplasticity. The results demonstrate that these active components significantly improve depressive-like behaviors and regulate key targets at the molecular level, including the 5-HT1A receptor, BDNF, and the NF-κB signaling pathway. Innovatively integrating multi-omics data with experimental validation, this study elucidates the multitarget synergistic mechanism of Chinese medicine's antidepressant effects, offering new insights into the modernization of traditional Chinese medicine research and laying a foundation for the development of highly efficient and low-toxicity antidepressant drugs.
