高效液相色谱法在药物分析中的应用与优化
摘 要
通过文献综述与实验研究相结合的方法,本文系统梳理了HPLC在药物成分检测、代谢产物分析及质量控制等方面的应用进展,重点考察了流动相组成、色谱柱选择、检测波长等关键因素对分离效果的影响。实验结果表明,采用梯度洗脱程序结合C18反相色谱柱可显著提高复杂药物样品的分离度;通过优化流动相pH值至5.0-6.0范围,可有效改善碱性药物的峰形对称性;同时,引入二极管阵列检测器(DAD)实现了多组分药物的同步检测。本研究的创新点在于提出了一种基于响应面法的HPLC参数优化策略,并通过实验验证了该方法的可靠性与实用性。研究结果为药物分析领域的HPLC方法开发提供了理论依据和实践指导,对提高药物质量控制的准确性和效率具有重要意义。

Abstract
Through the method of combining literature review and experimental research, this paper systematically reviewed the application progress of HPLC in drug composition detection, me tabolite analysis and quality control, and focuses on the influence of key factors such as mobile phase composition, column selection and detection wavelength on the separation effect. The experimental results show that the gradient elution procedure combined with C18 can significantly improve the separation; the peak symmetry of alkaline drug can be improved by optimizing the mobile phase pH to 5.0-6.0; meanwhile, the diode array detector (DAD) is introduced to realize the synchronous detection of multi-component drug. The innovative point of this study is to propose a HPLC parameter optimization strategy based on the response surface method, and to verify the reliability and practicability of this method through experiments. The results provide theoretical basis and practical guidance for the development of HPLC methods in the field of drug analysis, and have important implications for improving the accuracy and efficiency of drug quality control.
