糖皮质激素在免疫调节中的多途径作用机制研究
摘要
糖皮质激素作为一类关键的甾体激素,因其显著的抗炎、免疫调节及抗休克特性,在临床治疗中被广泛应用。然而,长期使用或大剂量应用GCs可能引发多种不良反应,如肥胖、高血压、骨质疏松等,严重限制了其临床应用的安全性和有效性。近年来,随着生物技术的飞速发展,对GCs作用机制的研究日益深入,但仍存在许多未解之谜。特别是在免疫调节方面,GCs的多途径作用机制尚未完全阐明,这阻碍了更精准、更安全的GCs类药物的研发与应用。因此,本研究旨在深入探讨糖皮质激素在免疫调节中的多途径作用机制。通过对GCs的药理作用、作用机制及其在免疫耐受与自身免疫病中的应用进行全面分析,本研究揭示了GCs通过基因表达调控、表观遗传学调控、细胞凋亡与存活平衡、炎症介质合成与释放及细胞因子网络调节等多种途径发挥免疫调节作用。这些发现不仅为理解GCs的生理和药理功能提供了新的视角,也为相关疾病的治疗提供了新的思路和方法。本研究对于优化GCs类药物的临床应用、减少不良反应、提高治疗效果具有重要的指导意义。

Abstract
As a key steroid hormone, glucocorticoids are widely used in clinical treatment due to their remarkable anti-inflammatory, immunomodulatory and anti-shock properties. However, long-term use or high-dose application of GCs may trigger multiple adverse effects, such as obesity, hypertension, and osteoporosis, which severely limits the safety and effectiveness of their clinical application. In recent years, with the rapid development of biotechnology, the action mechanism of GCs has been increasingly studied, but many unsolved questions remain. Especially in terms of immune regulation, the multipathway action mechanism of GCs has not been fully elucidated, which hinders the development and application of more accurate and safer GCs drugs. Therefore, this study aimed to deeply explore the multipathway action mechanism of glucocorticoids in immune regulation. Through the pharmacological effect of GCs and its application in immune tolerance and autoimmune disease, this study revealed the GCs through gene ex pression regulation, epigenetic regulation, apoptosis and survival balance, the synthesis and release of inflammatory mediators and cytokine network regulation of immune regulation role. These findings not only provide new perspectives for understanding the physiological and pharmacological functions of GCs, but also provide new ideas and approaches for the treatment of related diseases. This study has important guiding significance for optimizing the clinical application of GCs, reducing adverse reactions and improving therapeutic effects.
