摘 要
本文系统性地探讨了非甾体抗炎药物(NSAIDs)的合成策略及其复杂的抗炎作用机制,旨在为这一类药物的深入研究和临床应用提供全面的参考。在合成方法方面,本文详细介绍了芳基重排法(特别是以布洛芬合成为代表)、BHC绿色合成法、卤代烃羰基化法及烯烃羰基化法等前沿技术,这些方法的阐述不仅展示了NSAIDs合成的多样性和创新性,也反映了现代化学合成技术在药物研发中的重要作用。在抗炎机制研究方面,本文深入剖析了NSAIDs如何通过抑制环氧化酶活性来阻断炎症介质的合成与释放,特别是通过解析环氧化酶的结构与功能,以及化学结构-活性关系,阐明了NSAIDs在减少前列腺素等关键炎症因子方面的具体作用。此外,本文还探讨了NSAIDs在减少白细胞聚集与活化、调控免疫反应等多方面的抗炎机制,揭示了其多途径、多靶点的抗炎作用特点。最后,本文概述了NSAIDs在疼痛管理、炎症性疾病治疗、退热及外科手术后镇痛等临床领域的广泛应用,强调了其在现代医疗体系中的重要地位。

Abstract
In this paper, we systematically discuss the synthetic strategies of non-steroidal anti-inflammatory drugs (NSAIDs) and their complex anti-inflammatory mechanisms, aiming to provide a comprehensive reference for the further study and clinical application of this class of drugs. In terms of synthesis methods, this paper introduces in detail the advanced technologies such as aryl rearrangement (especially ibuprofen synthesis), BHC green synthesis, halogenated hydrocarbon carbonylation and olefin carbonylation. The desc ription of these methods not only shows the diversity and innovation of NSAIDs synthesis, but also reflects the important role of modern chemical synthesis technology in drug research and development. In terms of anti-inflammatory mechanism research, this paper in-depth analyzed how NSAIDs blocked the synthesis and release of inflammatory mediators by inhibiting the activity of cyclooxygenase, especially by analyzing the structure and function of cyclooxygenase and the relationship between chemical structure and activity, and clarified the specific role of NSAIDs in reducing prostaglandins and other key inflammatory factors. In addition, the anti-inflammatory mechanism of NSAIDs in reducing leukocyte aggregation and activation, regulating immune response and other aspects was also discussed, and the characteristics of its multi-pathway and multi-target anti-inflammatory effects were revealed. Finally, this paper summarizes the extensive application of NSAIDs in the clinical fields of pain management, inflammatory disease treatment, fever reduction and postoperative analgesia, and emphasizes its important position in modern medical system.
